Predicting Survival in Resected Pancreatic Cancer Using Gene Mutations

This study developed a survival prediction model integrating somatic driver gene mutations in patients with resected pancreatic ductal adenocarcinoma (PDAC), revealing that a higher number of mutations correlates with increased mortality risk.

  • In a cohort of 663 patients, the median overall survival (OS) was 28.4 months, with a significant increase in mortality associated with each additional mutated gene (HR 1.27).
  • The integrated model demonstrated better discrimination (c-index 0.690) compared to traditional AJCC staging (c-index 0.568), supporting its potential clinical utility.

Surgeons should consider the cumulative burden of driver mutations when discussing prognosis with patients post-resection, but further validation is needed before adopting this model in practice.
