Patients with resectable or borderline resectable pancreatic ductal adenocarcinoma (PDAC) who received neoadjuvant chemotherapy and radiation (total neoadjuvant therapy, TNT) or neoadjuvant chemotherapy only (NAC) had longer overall survival (OS) and disease-free survival (DFS) compared to those who underwent upfront surgery. In a study with 26 patients receiving TNT, 28 receiving NAC, and 168 undergoing upfront surgery, those in the neoadjuvant therapy groups had improved outcomes, suggesting a beneficial role for preoperative treatment in PDAC management.
